Same here. And I push my car over 3000 RPM almost daily. On 100% highway driving I get 450-475 depending on how fast I go. I noticed I got about 20 miles more per tank when I switched over to Mobile 1 synthetic 5W-30. Also, I always change the oil every 3k and put injector cleaner in at the same time.
Oh yeah, I'm bone stock 2000 SLE V6 except for TRD rims.
If you are getting really bad mileage I would look at the following:
Spark plugs and wires: When I replaced mine at 60K they really looked burned. After the change I got 35-40 more miles per tank. Next time I plan on changing the plugs (OEM Platinums) at 45k intervals.
Injector cleaner: consider using the off the shelf stuff first, but the dealer's cleaning can help a lot because it cleans out the throttle body too. It's a high pressure system that they attach directly to the engine so it bypasses the fuel line and doesn't get diluted by a tank of gas.
Fuel Filter: commonly overlooked, but if it is you will have a greater resistance to the flow of fuel, making the engine work harder.
I'm not really surprised that you people have seen better mileage by putting on different low-resistance mufflers (TRD, etc.). There is less back pressure on the exhaust system and therefore the engine has to work less hard. It is the same reason why you get an 2-3HP by putting on that muffler: less resistance to flow. Remember, our stock mufflers were made to keep little old women happy: NO NOISE = HIGH FLOW RESISTANCE.
Just my .02.
